How Do I Calibrate Periotron Professional For GCF?

Insert a blank PerioCol strip between the Periotron fluid meter sensors, and set the instrument to read zero (0) by turning the dial to the left or right. Discard the blank strip once set to zero.

Using a microliter syringe, which can accurately deliver 0.25 to 1.25 µl of fluid, Periotron Scores are related to volume (µl) as follows: The syringe is filled to capacity (1.25 - 5.0 µl, depending on the model of syringe selected) with a test fluid making sure that the loaded syringe contains no air bubbles. It makes little difference whether the test fluid is distilled water, saliva or serum; the results are essentially the same, except for water at the upper limits of the range. The plunger is carefully depressed to deliver 0.25 µl, which appears as a minute droplet at the end of the syringe tip. Without delay, touch the end of a PerioCol strip to the test fluid droplet. Notice that it is immediately absorbed onto the strip. Again without delay, place the strip between the Periotron sensors. A Periotron Score is displayed shortly. Repeat this step three (3) more times with this volume (0.25 µl) and record the average score.

Repeat the above step using volumes of 0.5 ul, 0.75 ul, 1.0 ul and 1.25 ul, and in each case record the average Periotron score.

Once scores have been obtained for all volumes, Periotron Professional will compute a standard curve relating the volume of fluid (µl) on the "x" axis to the Periotron Score on the "y" axis. Please note that calibration usually only needs to be set once. To check that the calibration has not or hardly changed with time, simply measure the volumes of two (or more) known samples.

Placing a strip containing an unknown volume of fluid (e.g. gingival crevice fluid; GCF) or periodontal pocket fluid; PPF) between the sensors, a Periotron Score is automatically determined. The volume of fluid (µl) is determined by interpolation from the standard calibration curve.
