How Do I Calibrate the Periotron Model 8000 for Use With Periotron Professional 3.x?

 
  Calibrating the Periotron
Calibration of the Periotron fluid meter is only necessary if conversion of Periotron Scores to volume and/or fluid thickness measurements are desired. Periotron Scores without such conversion can be used as an indication of disease severity or lack thereof. Gingival crevicular fluid (GCF) level is a good indicator of gingivitis-periodontis severity. As a consequence, Periotron Scores can be used as an indicator of success or failure of patient treatment.

Sometimes, it is desirable to know the volume of a sample of saliva collected as for example when collecting saliva from a single minor salivary gland; other times it is desirable to know the thickness of a saliva film. To change Periotron Scores into volumes (µl) or into thicknesses (µm), a standard curve must be prepared. When SialoPaper is selected, dividing volume values from this standard curve by the area of a SialoPaper strip converts Periotron Scores into thicknesses in micrometers.

1. Click System/Calibrate
2. To calibrate for GCF, click PerioCol Paper. To calibrate for saliva, click SialoPaper.
3. Click the command button, Calibrate. Please note that if the Periotron has already been calibrated for either paper, the command button will be labeled Recalibrate.
 
  Calibrating the Periotron Standard Curves
A standard calibration curve is obtained by using a 4th order polynomial to plot the relationship between Periotron Scores and fluid volume. By taking measurements of known volumes at various intervals and recording the average Periotron Score at each interval, coefficients can be calculated which will convert Periotron Scores to sample volumes. In measuring saliva thickness, dividing the volume by the area of a SialoPaper strip yields the thickness of the saliva sampled. Coefficients are displayed at the bottom of the screen.

Once average Periotron Scores are obtained, enter the score in the box corresponding to the microliter volume of the sample. Click on Calibrate to have Periotron Professional calculate the coefficients and plot a standard calibration curve. It is important that calibration be done as accurately as possible as it will affect the accuracy of Periotron Score conversions to microliters (µl) and in turn to micrometers (µm).

Click on Done to save the calibration data, including coefficients, and return to the main menu. Clicking Cancel will ignore all entered scores and exit without saving any calibration data. Please note that the Periotron as a result will remain uncalibrated.
